Can i hook up usb seagate 4tb to usb port and use it, without an internal hard-drive? (i don't want to buy internal hard drive). thanks for replies

Why would you do such a thing?.... If you don't intend to use it what's it made for then don't buy it. You can plug your hard drive to your router's usb port if you have one and use it at network drive but with very limited options.

Is there a limit to the size of hard drive that can be used

This Bundle comes with 4 x 4TB Drive but the NAS supports upto 22TB Drives on each bay.

can this work as a media server like my twonky shows on my network 4k disk player menu?

Yes QNAP has DLNA Server built in to the NAS. Please install below Apps from App Center to enable DLNA, Multimedia Console Media Streaming Add-on

Has anyone used this as a replication target from another QNAP?

Yes this can be used as a Replication Target any another QNAP. You can do Real-Time Sync of your Data from another QNAP or Schedule a Replication Job from another QNAP.
