What is the measurement of space between each of the prongs? There seems to be a few kinds with different spacing.

The one I have has 4 holes. Two of the holes are spaced about 1/2" from both edges of the paper and the other two holes are 3" in from the center of the first edge holes (or 3 1/2" in from the edge). I hope this helps.

Does this binding system need a machine to complete the process or can it be done by hand?

That part gets done by hand, but a machine is needed to punch the holes, the holes are smaller than the traditional hole puncher makes and it makes 4 holes instead of 3.
